Angiogenesis, the formation of new blood vessels, is thought to play an important role in the establishment and propagation of primary as well as secondary metastatic tumors (Weidner, N., et al.  N. Eng. J. of Med. 324:1-8. 1991)

Several synthetic and naturally occurring inhibitors of angiogenesis have been described. Experimental synthetic inhibitors of angiogenesis such as minocycline, a-difluoromethylornithine, and analogs of heparin and fumagilin have shown promising results in vitro and in some in vivo studies.  Naturally occurring inhibitors of angiogenesis such as thrombospondin and platelet factor 4, both isolated from blood platelets as well as tissue inhibitor of metallproteinases (TIMP), purified from mesenchymal cells derived from cartilage, have demonstrated potent anti-angiogenic activity as well as anti-proliferative activity for vascular endothelial cells (Passaniti, A., et. al. Lab. Invest. 67:519-528. 1992)

More recently, naturally occurring, putative angiogenesis inhibitors including Angiostatin and Endostatin have been described (O'Reilly, M.S., et. al. Cell 79:315-328. 1994 and O'Reilly, M.S., et. al. Cell 88:277-285. 1997).
