Description of Services and Deliverables
Phase I
Client needs to provide 5 frozen aliquots of 250 µg antigen in 500 ml PBS each (500 µg/ml).
Immunization of 5 Balb/c mice with specific immunogen.
Tail bleeds tested for specific antibody.

Phase II
Fusion of spleen cells with P3x63-AG8.653 myeloma cells.
Screening culture plates for specific antibody (ELISA).
Selection and expansion of up to 25 positive fusion products
.
Media supernatant from expanded cells shipped to customer for additional testing.

Phase III
Subcloning of up to 5 selected clones per fusion to monoclonality by limiting dilution.
Cryopreserving 10 to 12 vials per selected subclone.

...the myeloma line is deficient in one of the essential salvage pathways for DNA synthesis and so does not survive when the main biosynthetic pathway for DNA synthesis is blocked. The unfused spleen derived lymphocytes do not survive because they are physiologically programmed to die unless “rescued”. 

The cells which survive in the appropriate medium are hybridomas, the myeloma-spleen cell hybrids resulting from the fusion. The hybridoma derives its immortality and antibody secreting capacity from the myeloma cell genes, and the salvage pathway genes and antibody-coding genes from the spleen B lymphocyte.
