Routine negative staining for either TEM or SEM includes processing of the samples, application on to grids, microscopic analysis of surface morphology, and photographic documentation. Quantitative negative staining also involves the counting of particles, including viruses. Statistical analysis is also performed.
Immunogold Labeling 70030-00
Immunogold labeling is a method used for the localization of specific antigens at the EM level. Samples are fixed (using lower concentrations of glutaraldehyde), processed, and sectioned. The grids are immunostained using an antigen-specific primary antibody followed by a gold-labeled secondary antibody. Electron-dense gold particles localize to the desired antigens within ultastructural components and organelles of cells.
