2D Gel Electrophoresis with Characterization of Proteins by Western Immonblot using a Single Primary Antibody:

  • After performing 2-D electrophoresis as described above, the unstained resolved gel is transferred to PVDF membranes.
  • Transferred proteins (spots) are then visualized by fluorescence staining, imaged and stored as tif files.
  • The characterization of individual spots is then performed by Western Immunostaining of the same blot. A specific (primary) antibody is followed by a biotinylated secondary antibody and the complex is detected with Streptavidin HRP or ALK Phos using the appropriate substrates or Protein A Gold conjugates.
  • Sequential probing with several primary antibodies (up to 3) can also be performed.
  • Digital images of the blot are then superimposed on the fluorescent image previously captured to demonstrate the IEF and Molecular Weight location of a specific spot. Client would receive digital images of the resolved blot and immuno-detected spots.

  • Following sample preparation, electrophoresis and staining of gels, unique spots will be identified and x/y annotated using Compugen’s Z3 software.

  • Experimental data is then matched to existing databases in pursuit of identification of a protein.
