High Throughput Protein Purification

Affinity tagged recombinant proteins are expressed and purified (~ >90% pure) using a one-step purification procedure from crude lysates or cell culture supernatants. Briefly, cells are lysed under native or denaturing conditions, (membrane proteins, inclusion bodies, etc) applied to 96 well plates, and incubated with metal-chelate affinity resin. The resin is washed to remove nonspecifically bound contaminants, and the protein of interest is eluted using either increased imidazole concentrations or low pH.

The process typically yields 1-50 µg purified protein. Alternatively, increased protein yields can be obtained using affinity spin columns or larger plates. The capacity of Ni affinity resin is reported to be in the range of 5-10mg/ml.
 
In addition, using Ni-NTA magnetic agarose beads (Qiagen), affinity purified recombinant protein can be assayed in 96 well format prior to elution to study biomolecular interactions (ie. protein-protein, DNA-protein, immunoassay)
