Antibody Discovery

Paragon’s current program for therapeutic antibody generation is based on traditional hybridoma generation as well as current display technologies. Display technologies result in fully human antibody products.

Display technologies

The screening of Phage Display or Yeast Display libraries will be subcontracted. Library screening (panning) will be performed by subcontractor. High-Throughput screening (HTS), Solution-based and Cell-based selection will be subcontracted or performed at Paragon. Transfection (generation of antibody product), DNA sequencing, and analysis will be subcontracted. Subsequent screening for binding and functionality will be performed by Paragon.

Such screening will include:
ELISA
Western blot
flow cytometry
Immunofluorescence microscopy
BIACore off-rate screening

 

Traditional Hybridoma

In general, the process includes immunization of an animal to generate an antibody response, screening, selection and immortalization of specific antibody-secreting cells, and subcloning the desired cells to ensure stability and monoclonality.

Selected mAbs are screened for:
Binding (e.g. ELISA, Western blot)
Affinity (e.g. BIACore, Forte Octet)
Functionality (e.g. FACS, neutralization)
