Large-Scale Purification of Recombinant Viruses,
Adenovirus & AAV's, and Virus-like Particles
 

Development of a standard operating procedure for the large-scale purification of recombinant viruses can be performed using large-scale ultracentrifuge (PKII ultracentrifuge). Protocol based on continuous flow operation with isopycnic banding in an appropriate gradient material. Choice of gradient material, process flow rate and banding time determined for each virus based on biophysical characteristics (size, density and sedimentation coefficient) and analytical experiments. All procedures performed under GLP compliance and the developed process will be both scaleable and able to be validated for future GMP manufacturing.
	The following deliverables will be provided to the customer upon completion of the project: 1. Concentrated virus dialyzed into suitable buffer 2. Specification sheet including: Density profile and ELISA of recovered fractions  Viral titer by TCID50 assay Morphology by EM (negative staining) SDS-PAGE/Western blot  3. Purification SOP
